적혈구 모양이 낫모양이 됨으로, 빈혈증세를 일으킴. 그러나, 이 원인 유전자가 NoSmoke:말라리아 에 저항성이 있음으로써, 인간유전자풀에 남아있게 된다. 

HemoGlobin의 beta-unit(beta-[[Globin]])의 6번째 AminoAcid가 GlutamicAcid에서 [[Valine]]으로 치환됨으로써 일어난다. 산소가 결합하면 HemoGlobin들이 서로 결합함으로써 한쪽으로 치우친 모양(낫모양)으로 변한다.
