ProteomeResearchNewFrontiersInFunctionalGenomics이후 그럴듯한 [Proteomics]관련 책이 없었다. 일단 이책을 통해서, 최근의 [Proteomics]에 대해 제대로 이해할 수 있을 것이다.
서문에 소개되는 DnaMicroArray만으로는 한계가 있는 이유 (즉 [Proteomics]가 중요한이유)
- mRNA abundance does not always correlate well with protein abundance.
- The sensitivity and dynamic range of existing approaches are such that the lowest abundance mRNAs (potentially encoding the most important regulatory proteins) are not readily measured alongside the more abundant mRNAs.
The activity of the proteins encoded by mRNAs is regulated at several levels beyond their mRNA or protein expression by, for example, their subcellular localization and/or the extent to which they are in PostTranslationalModification, neither of which are revealed by measurement of mRNA abundance.
- There are a number of important biological samples, particularly those that might be used for human diagnostics, such as urine, cerebrospinal fluid and blood plasma, that do not contain mRNA.
- The analysis of mRNA expression in human biopsy and post-mortem samples is still a significant challenge given the potentially protracted time between the collection of the sample and the vulnerability of mRNA to degration.
DNA sequence based approaches will have to be complemented by the direct analysis of the products encoded by the genes - the proteins. for example, although genome sequences can be used to predict OpenReadingFrame, such predictions are not infallible.
